THE ESEM E-BOOK

We have produced an electronic-handbook for Environmental Scanning Electron Microscope (ESEM) users in Life Sciences. As this technology is not widely used in Singapore, the handbook can be used as a convenient form of reference for the current and future users of the ESEM.

The specimens used are fungi of different species and their morphology differences were taken note of using the Environmental SEM. The variables in the experiment are the different ways of specimen preparation and the different viewing conditions such as the pressure and the accelerating voltage used. To compare, another batch of fungi underwent normal processing and viewing under the conventional high-vacuum SEMs. Over 400 digital images were taken and selected ones are featured in the handbook.

The handbook contains:

1) Specimen preparation techniques for ESEM
2) Optimised ESEM conditions for imaging
3) Comparisons of image quality with conventional SEMs
4) The "Dos and Don'ts"
